Start with the product profile and clinical question

The team defines the target, mechanism, proposed indication, population, route and likely regimen. These choices shape a product profile and a map of critical uncertainties.

An oncology product may require a different risk balance, model selection and nonclinical package from a product intended for long-term use in a healthy population. The plan must be product- and context-specific.

Pharmacology and exposure-response

Primary and secondary pharmacology explore intended activity and possible off-target effects. ADME and pharmacokinetics describe absorption, distribution, metabolism and excretion and connect dose to systemic exposure.

A model is useful only to the extent that it is relevant to people and the clinical question. An in-vitro result does not guarantee a reproducible in-vivo effect or an acceptable risk profile.

Safety and GLP

Toxicology evaluates potential target organs, dose- and exposure-related effects, reversibility and parameters for clinical monitoring. Specified regulatory safety studies may be conducted within GLP systems that preserve study plans, procedures, raw data, quality assurance and reports.

ICH M3(R2) connects the duration and nature of nonclinical studies to the proposed clinical programme. ICH S9 provides principles specific to anticancer pharmaceuticals and their development context.

Manufacturing and analytics develop in parallel

Biological findings cannot be separated from the material tested. Composition, impurities, stability, analytical methods and batch comparability influence both safety and the transferability of conclusions.

As the programme approaches the clinic, nonclinical work must align with CMC and the manufacture of clinical material. Poor alignment can make earlier studies difficult to interpret or require repetition.

The decision to enter clinical development

The integrated assessment brings together mechanism, pharmacology, exposure, toxicology, product quality, clinical need and risk management. It supports the proposed starting dose, escalation, monitoring and stopping criteria.

Expert review is not a promise of success. Its value is finding critical gaps early and creating a coherent package for regulatory, ethics and clinical discussion.

Quick answers

Frequently asked questions

Are preclinical and nonclinical studies the same?

The terms overlap in ordinary use. International guidelines commonly use nonclinical; the precise meaning depends on the document and stage of development.

Must every preclinical study follow GLP?

No. Applicability depends on the study purpose and regulatory role. Discovery and mechanistic studies may use other systems while still requiring scientific integrity and traceability.

Can animal data predict efficacy in people?

Only to a limited extent. Such data can inform mechanism, exposure and risk, while clinical efficacy must be established in research involving people.
